2023년 6월 온라인 세미나 소식 알려드립니다.

이번에 모실 연사는 Stanford University의 Possu Huang 교수님입니다. Huang 교수님은 2010년대 초반 de novo protein design 분야를 새롭게 개척한 핵심 인물 중 한 분으로, Baker lab에서 postdoc 이후 Stanford bioengineering department에 교수로 현재까지 재직 중입니다. 최근에는 딥러닝을 이용한 Antibody design 관련하여 활발히 연구하고 계시기에 이번에 특별히 초청하여 말씀을 듣고자 합니다.

제목: Protein Design for Epitope-specific Molecular Recognition

The growing need for antibodies with customized specificity provides a rich environment for engineering efforts. In recent years, despite having streamlined experimental pipelines, the fundamental math requiring extensive libraries and screen campaigns to get an initial binding signal remains unchanged. A major advancement would be to directly design an in silico epitope-specific binder from scratch, providing a signal for potential optimization by artificial evolution. We have observed several key advantages in neural network approaches over existing methods. By leveraging the unique properties of generative neural networks, we developed a model for immunoglobulin 3D structures, with which diverse structures can be modeled with unprecedented speed. We extended it to a purely deep learning-based protein-protein interface design pipeline that optimize not only spatial orientations but fully-flexible protein structures on the fly to desired epitopes. This novel strategy explores neural network’s capabilities in modeling dynamic structures, and preliminary experimental results on multiple targets support the plausibility of in silico design of epitope-specific antibodies.
